Description:
Given the vital importance of immune system research, the gathering of clear, consistent, and informative protocols involving the study of dendritic cells is paramount. Bringing the popular first edition fully up to date, Dendritic Cell Protocols, Second Edition presents protocols from experts in the field that cover the basics and more complex forays into the exploration of DC development and function, both in mice and humans. The first section of the volume involving humans explores topics such as the isolation of blood DC subtypes, primary skin Langerhans cells, and the generation of gene-manipulated human DCs with the inclusion of more clinically relevant methods as well, while the second section involving rodent models delves into DC and precursor generation in vitro, isolation ex vivo, disease models, as well as DC functions and properties. Written in the highly successful Methods in Molecular Biologyв„ў series style, chapters include introductions to their respective subjects, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and notes on troubleshooting and avoiding known pitfalls. Comprehensive and cutting-edge, Dendritic Cell Protocols, Second Edition aims to become a bench-side handbook for both beginners and experts in the field of DC research and a long-term reference for some of the most popular methods put forward by those who lead the field. Description:
Pulling a handbag from a pushchair handle, two teenage trouble-makers, Zipp and Andreas, unwittingly commit murder when the baby inside is thrown to the ground. Unaware of what has happened, the pair move on to their next target. Having followed home Irma, an elderly lady who lives nearby, Andreas enters her house armed, with his trusty flick-knife. Zipp waits nervously outside, but his friend never reappears. He will never see him alive again. Fossum toys with the roles of victim and killer and pens the inner monologue of her characters to chilling effect. There are no red herrings in this narrative as the crime is played out on the page, but it is the mental processes behind the murderer's sinister act that are the real pieces of this puzzle. Gradually, they fall into place as Irma and Andreas talk to each other. We are forced to question the familiar stereotypes: people are not always what they seem and it is not always bad people who do bad things. While the reader knows the truth of this crime, the police remain flummoxed. There is no reason for Sejer and his colleague Skarre to see a connection between the infant's death and the reported disappearance of a town trouble-maker.

Description:
In recent years, our ability to understand and manipulate epidermal cells has increased tremendously, opening significant new possibilities in both basic science research and in regenerative medicine, including would healing and transplantation. Epidermal Cells: Methods and Protocols, Second Edition expands upon the popular first edition by bringing together a panel of experienced basic and clinical researchers to describe in step-by-step detail the powerful methods they have developed and optimized to analyze and manipulate epidermal cell precursors and mature epidermal cells. These protocols cover different methods and models for culturing epidermal cells, for enriching very early epidermal progenitors, as well as for studying epidermal cell commitment and differentiation both in vitro and in vivo. Topics of special interest include the derivation, characterization, and utility of epidermal stem cells, mature epidermal cells and their characterization, and applications in regenerative medicine. The protocols follow the successful Methods in Molecular Biologyв„ў series format, providing introductions to their respective topics, lists of the necessary materials and reagents, and notes on troubleshooting and avoiding known pitfalls. State-of-the-art and highly practical, Epidermal Cells: Methods and Protocols, Second Edition offers experienced and novice investigators alike an invaluable collection of readily reproducible techniques designed to broaden not only our understanding of the biology of epidermal cells, but also their utility in normal tissue homeostasis and regenerative medicine applications.
